The defending champions, who have only dropped two points in the top-flight this season, went ahead courtesy of Rodrigo Bentancur's fine finish in the 31st minute.
Captain Giorgio Chiellini doubled their lead after the interval in scrappy fashion before Cristiano Ronaldo added gloss to the scoreline from the penalty spot.
The result emphatically ended Fiorentina's run of five successive draws as Juventus showed no sign of a European hangover.

Juventus win here in Florence, and were very impressive throughout. Rodrigo Bentancur opened the scoring after half an hour, and a quick-fire two goals from Giorgio Chiellini and Cristiano Ronaldo in the second-half sealed it for Juve, who are now 11 points clear at the top of Serie A.
They're heading for an unprecedented eighth straight Scudetto.
For Fiorentina, the loss was their first at home this season, and they now haven't won in seven games. They next travel to Sassulo next Sunday.
Juventus have less than a week to prepare for a massive clash on Friday, as they welcome Inter Milan to Turin.
