The 20-year-old, who was the first Japanese woman to appear in a Grand Slam final, was barely 12 months old when Williams won her first major title but overcame the legendary Williams in straight sets 6-2 6-4 to win at Flushing Meadows.
Williams, meanwhile, attracted headlines for all the wrong reasons after on-court violations and a row with the umpire.
The 36-year-old had the chance to draw level with Margaret Court's all-time record (24) of Grand Slam titles but fell to the youthful energy and classy all-round play of Osaka.
Here is how it happened:

Williams 1-0 Osaka*
Serena starts with a fault, a let and then a long backhand which misses the baseline, giving Osaka her first point in a Grand Slam final. And she gets her second with a double fault.
But just like that the slight opening flicked away with a hammer-blow of a forehand dow the line and then a couple of big serves and she takes it on deuce.
Serena is no stranger to slow starts in her career so try not to read too much into this.
Williams* 1-1 Osaka
An unforced error and a double fault and Osaka starts her first service game in identical fashion to her idol at the other end of the court here on Arthur Ashe.
And, just like her idol, Osaka fights back with some heavy forehands and levels the set. Good strength of character there from the 20-year-old.
Williams 1-2 Osaka*
Serena is just going through the motions here but again Osaka is fighting back and forces her opponent into another error after a great rally. And a long forehand sees Osaka win the first break point of the match.
And a double fault from Serena sees the Japanese break the American! That's not what was supposed to happen! What a start from Osaka.
Williams* 1-3 Osaka
So she had the break... but can she consolidate it? A great, low backhand says she can, a perfectly judged forehand says she will and an ace confirms she has! Nerves? What nerves?
Williams 1-4 Osaka*
Don't, not that you ever, EVER, would, rule out Serena. She starts with two big points but again Osaka has the answer and then there is a third double fault already for the home favourite. And another long forehand and Osaka has yet another break point!
And she takes it! Osaka has the double break and this is a very slow start from Serena! Anyone predict this?
Williams* 1-5 Osaka
This is a huge game in this match now. WOW! What a stunning cross-court pass from Osaka, even Serena acknowledges it! But a huge scream from Williams in the following point and you get the feeling she is starting to feel this a bit now. She's trying to go big on ever groundstroke now.
And it's working! Serena now has a break point. But she can't take it! Osaka aces but follows it up with an unforced error. another break point. And another save. That is the 18th break point Osaka has saved in a row - she hasn't been broken in 22 service games now.
Williams 2-5 Osaka*
Serena is rattled. Another double fault and Osaka could take the first set in less than half an hour. But she can't as Serena comes back from 0-30 down to take the game! Osaka to serve for the set next...
