Mediate, who never led outright until the final stroke of the tournament, scored a birdie at the final hole to record a six-under-par 66 in his final round to finish at 20-under with a total of 268.
After Mediate sunk his 12-foot putt, his fellow American Chris Perry pushed his eight-foot par putt left of the hole to finish with a bogey and lose the title by the narrowest of margins.
Hal Sutton was three strokes back in third, with Phil Mickelson and Woody Austin sharing fourth at 273 - one stroke ahead of five others.
The American Meg Mallon shot a final round three-under-par 69 to win the du Maurier Classic at Royal Ottawa yesterday - but the final Major of the season turned out to be desperately disappointing for the Britons.
Mhairi McKay was the best of the British players in joint-13th place on four over par at 292 - but she would have been 11th and around £1,200 richer if she had not been given a two-shot penalty for slow play at the 6th hole that turned a 72 into a 74.
England's Trish Johnson, fifth overnight, slumped to a final-round 79 and out of the top 10, while Laura Davies finished last of the qualifiers on a 23-over-par 311.
