The Argentina No 1 damaged his right knee in a training session and immediate tests from the team doctors suggested Romero will miss five-to-six weeks.
It is a blow for Jorge Sampaoli, who stuck by the 31-year-old Romero even as he struggled for regular playing time with United this season.
Nahuel Guzman, who was included in Argentina’s longlist but axed when the squad went down to 23 this week, is expected to replace him.
However Guzman’s father strongly criticised Sampaoli for leaving his son out, posting insulting messages about the Argentina head coach on social media.
River Plate’s Franco Armani or, most likely, Manchester City’s Willy Caballero will replace Romero as first-choice keeper for this summer’s tournament.
