Jose Mourinho takes charge of Spurs for the first time as they visit West Ham, with Tottenham's first team in the post-Mauricio Pochettino era sure to be fascinating.
Arsenal look to stave off a crisis of their own against struggling Southampton, and Liverpool travel to Selhurst Park to take on Roy Hodgson's Crystal Palace.
Saturday evening sees Manchester City host Chelsea looking to go back ahead of Frank Lampard's side, with Pep Guardiola keen to right the wrongs of the defeat to Liverpool.
And finally Ole Gunnar Solskjaer, perhaps looking over his shoulder with Pochettino now readily available, takes his Manchester United side to Bramall Lane for a tricky meeting with Sheffield United. Here’s how we think the Premier League’s big six might line up this weekend:
Tottenham (vs West Ham)
Jose Mourinho's first Tottenham Hotspur lineup will be utterly fascinating, and could offer some clues about how he perceives certain members of the Spurs squad.
It is somewhat difficult to predict exactly how he will assemble his first side, though it will likely be his preferred 4-2-3-1, and knocks to Tanguy Ndombele, Jan Vertonghen and Erik Lamela clarify things further.
After his high-profile promise to invest in the club's youngsters, we are tipping Juan Foyth and Kyle Walker-Peters to start. Eric Dier will likely be moved up into midfield, with Lucas and Son Heung-min selected out wide.
Arsenal (vs Southampton)
Arsenal gambled on a five-at-the-back formation last time out, and there are merits to persisting with such a system in the short-term, at least.
The personnel may change, however, against struggling Southampton, with Unai Emery potentially unleashing a potent front three of Pierre-Emerick Aubameyang, Alexandre Lacazette and Nicolas Pepe, with Mesut Ozil perhaps dropping out.
Granit Xhaka is not likely to return to the side after being stripped of the captaincy, but Kieran Tierney and Sokratis may be brought back into the side.
Liverpool (vs Crystal Palace)
With a big clash with Napoli looming, Jurgen Klopp could look to rotate his midfield against Crystal Palace, particularly with Jordan Henderson struggling with an illness recently.
Both Mohamed Salah and Andrew Robertson withdrew from international commitments due to injury, but could well be fit to start at Selhurst Park.
Naby Keita may get an opportunity in midfield.
Manchester City (vs Chelsea)
Pep Guardiola gambled with his formation and tactical set-up against Liverpool, but is likely to select a more traditional lineup in another big game against Chelsea.
Rodri returned from injury last time out and should keep his place, while Benjamin Mendy could replace Angelino at left-back.
David Silva should be fit for a midfield return.
Chelsea (vs Manchester City)
Frank Lampard will once again be able to call upon Jorginho with the midfielder serving a one-game suspension last time out.
That means Lampard may select an ostensibly first-choice midfield of N'Golo Kante, Jorginho and Mason Mount as they look to combat Manchester City's prowess in that area.
The key decisions perhaps come at full-back - Emerson may be preferred to Marcos Alonso, and while Reece James has impressed recently, Cesar Azpilicueta's experience and perceived solidity may be preferred at the Etihad Stadium.
Manchester United (vs Sheffield United)
Manchester United visit Bramall Lane with a squad returning to fitness, with Luke Shaw, Axel Tuanzebe and Nemanja Matic among those back in training.
Scott McTominay is a doubt after picking up an injury during the international break, but should be fit to start against Sheffield United.
It seems likely that Shaw and Tuanzebe are eased back in after their injury layoffs, which means Victor Lindelof and promising left-back Brandon Williams could retain their places in the back four.
