The 30-year-old club captain's place in the side has been questioned in recent weeks following several sub-par performances.
Rooney is one of four to drop out of Mourinho's starting line-up following last weekend's defeat to Watford.
Marouane Fellaini, Luke Shaw and Anthony Martial also fall out of the side, though the latter two ended the 4-2 reverse at Vicarage Road with groin and concussion injuries respectively.
Daley Blind comes in at left-back, Ander Herrera starts in midfield, while Jesse Lingard and Juan Mata come into the forward line.
Rooney, who played up front in Wednesday's 3-1 EFL Cup win at Northampton Town, is among the substitutes.
Leicester name an unchanged side from last weekend's 3-0 win over Burnley.
Islam Slimani, who bagged a brace on his home debut for the Foxes, plays alongside Jamie Vardy up front.
