The match at Old Trafford is the only Premier League game of a weekend dominated by the FA Cup, and having already drawn at Manchester City this season Brown insists the same can be done against United.
Visits to Manchester have dominated Hull's continuing rollercoaster ride in the Premier League - a thrilling 4-3 defeat at Old Trafford last season was followed by the infamous half-time team talk on the pitch at Manchester City.
This season Jimmy Bullard inspired a 1-1 draw at Eastlands and though he will be missing against Sir Alex Ferguson's side, Brown says his players - brimming with confidence after a goalless draw at Tottenham - can capitalise on a United side who are not in the best form with just one win in their last four games.
Brown said: "We have to take the same togetherness, the same spirit and the same mentality that we had at White Hart Lane last week.
"If we bring that to the table, I think there is a great opportunity for a result to follow.
"There's references from last season for us having taken United so close at Old Trafford and scoring three goals there.
"But I'm not sure there's any such thing as a good time to play Manchester United."
Keeper Boaz Myhill was the hero last weekend with a string of incredible saves to deny Spurs but Brown insists it is wrong to single out a single player.
He added: "A lot of references were made to Boaz Myhill's performance against Spurs, but I felt the clean sheet was down to the spirit and the togetherness of the squad.
"We've got a magnificent changing room full of character and experience. That all came to the party last week and something similar has to happen on Saturday.
"Old Trafford is an exciting place when the game is flowing and I've no doubt that will happen this weekend.
"I'd like to see a bit more of a cutting edge from us than we saw at Tottenham, but we have to be at our best defensively first and foremost."
Brown confirmed that loan signing Amr Zaki, the former Wigan striker, will not feature tomorrow due to a lack of match fitness.
"Hopefully Amr will bring goals. He did that with Wigan last season before going back to Zamalek," said the Hull manager.
"He's had a bit of a stop-start season with Zamalek, but hopefully with the regime we have here we can get him on the training ground doing double sessions and get his fitness levels up for Premier League football.
"This weekend would be a bridge too far for him, but with a good week of training under his belt, he will hopefully play some part against Wolves."
