Brook, 28, from Sheffield, is in a "stable condition", according to his promoters Matchroom.
The altercation is believed to have taken place this morning at 6am, and the boxer suffered the injuries to his left leg and a cut to one arm. No arrests have been made.
A spokeswoman for Spain's Guardia Civil said Brook was stabbed in the left leg at Golf del Sur in San Miguel de Abona and "lost a lot of blood".
She told the Press Association: "This morning a 28-year-old British man was stabbed at Golf del Sur in San Miguel de Abona.
"He was stabbed in the left leg and is hospitalized. He lost a lot of blood.
"There are no arrests at this time and the Guardia Civil are investigating."
Brook won his first world title last month after he defeated Shawn Porter via a majority decision in California to claim the welterweight strap.
The incident follows former European champion Jamie Moore being shot in Marbella in August.
Speaking to Sky Sports News, Moore said: "It's shocking to be honest. All I can say is I hope for a safe recovery. It's a bit scary to be honest, I just hope he is ok."
