The unnamed man, in his thirties, was detained after cold case officers opened up the investigation into the death of Rikki Neave.
Rikki, from Peterborough, was strangled to death in 1994 after last being seen heading to school on November 28 that year.
The schoolboy’s body was found naked in nearby woods the next day.
The post-mortem concluded he had been strangled.
Rikki’s mother, Ruth Neave, was charged with his murder, but a jury later found her not guilty.
She did plead guilty to child neglect and cruelty and was sentenced to seven years behind bars.
In recent years, she called on police to re-open the case, maintaining her son’s killer remains at large.
The 45-year-old previously said: 'I loved Rikki, he was a wonderful child.
“For the last 20 years, I have been going through living hell from the public opinion of me caused by all the lies.
“I wasn't a perfect mother but I never hurt my children and I should never have admitted to those offences.
“All I want now is justice and for his killers to be caught.
“I know people out there think I'm a murderer but I'm determined to prove that somebody else did this to my boy."
The man, also from Peterborough, has been bailed until June 15.
