John King was rushed to hospital suffering serious head injuries after plunging from the black Hyundai Tucson.
The 18-year-old, of Littleport village, Cambridgeshire, was pronounced dead on Wednesday.
Investigators say the vehicle, which contained at least two other people, had stopped at the scene of the incident at 9.40am on Tuesday.
In a statement Cambridgeshire Police said: “A teenager who fell from a moving vehicle on the A10 in Ely has died.
“Emergency services were called to the incident at about 9.40am on Tuesday after reports the young man had fallen from a black Hyundai Tucson.
“John King, 18, of Littleport, was taken to Addenbrooke’s Hospital in Cambridge but sadly died on Wednesday.”
It asked anyone who witnessed the incident to call the force’s roads policing unit on 101 quoting incident 133 of July 30.
