Schumer, who was presented her award for Trailblazer of the Year by Jennifer Saunders, said: "I'm not nervous."
She added: "I'm very glamorous. I don't feel out of place here."
Schumer went on to explain: "I'm probably like 160lbs right now and I can catch a dick any time I want."
The comic has been known for making strong statements with her comedy, including her sketch The Last F**kable Day, which pokes fun at ageism in Hollywood; and Girl You Don't Need Makeup, a painfully funny One Direction music video parody.
She also parodies the way some female actors present themselves on talk-shows.
Schumer's sketches are showcased on her US programme Inside Amy Schumer.
"It will be very exciting when what I am doing isn't trailblazing anymore," she said, "but I'm very happy to have gotten this."
Having advised women to "definitely get vaccinated for HPV", Schumer added that she had one goal for the night.
And that goal was "to take my underwear off at the end of the night and not make it look like I blew my nose on it."
